Definitely a queen, probably camponotus pennsylvanicus.

Yep. Probably one of the sugar ant or carpenter ant species.

So, even though she has wings is she ready for a test tube and laying eggs? I found her on the pavement just crawling around.
Also I updated the Google photos Link in my last post. I'm a little bit worried that she's going to get out as she is working that cotton over and tugging it out of the way.
Should I be concerned?

Even thought she still has wings on there is a good chance she has mated and if you saw her crawling around she has most probably mated. You should not be worried about her escaping you put a lot of cotton.
