Lasius niger queens

How many Lasius niger ants queens can be in one colony? I've cought like 40 of them today.

Only one per colony. 40 ants is an excessive number of queens to catch. We should try to catch no more than 5.

If I put 6 queens in 1 test tube they will kill each other?

Yes. Shortly after workers come around, only one queen will remain.

I released about 40 and only kept 6, but I only have 1 test tube so I guess I'll just keep one and release other 5 . I thought I would be able to make a super colony with like 10 queens lol.

Only some species of ants have more then one queen. And lasius is not one of them.
Myrmica Rubra will have more then one but they fly in August to September and they sting so think very carefully before you make that choice haha.
Research is important before during and even after you have established a colony. There is always time to learn and to listen to others experiences. Live by this and your ants will thrive. Fail to do so and your experience may be brief.
