if the test tube is plastic i would bang it on something hard with the entrance pointing down and the cotton will slowly slide of. also putting water in it might help

I had a similar issue when I was getting started. I put water at the bottom of the test tube on the cotton and was able to hold the tube at the round(stuck cotton) side and flick it Manny times almost like throwing a yoyo. It slowly began to move and eventually came shooting out!
