You don't do it -- you let the colony do it when it is ready.

Are you saying you want to move the queen out of a vertical nest, to a test tube? And if so, why?

Ant Colonies should live in a test tube until they reach 20-50 workers. Place the test tube in an outworld and once the colony is big enough connect the nest and let them move in on their own.
