In such a case you should think about a "forceful" move. Basically make them move out since they don't seem to worry at all. ^^'
Doing some proper research is the best way to start an antcolony.

New to ant keeping, keeping a Messor structor colony. Also I have 2 camponotus queens and what seems, 2 tetramorium queens

Not really. I shake them into the new testtube but I use a small bottle that I cleaned thoroughly to avoid issues. It plugs up testtube A, I shake some in. I plug it to testtube B, I shake them out. Sometimes Eggs still get stuck but those I move with some cotton or my tweezers.
