I just want to ask if its okay to put cotton in the disc shape hydration chamber under the ant tower

I would just break it up in small pieces.
Keep:
3 camponotus pennsylvanicus
2 formica subsericea

I think you misunderstood? The space is disk-shape, not the cotton he wants to put in? And yes that would be fine.

I meant break the cotton up and put it into the tray.
