I have a new container that I put mesh in for ventilation and got into stay with hot glue

I never made a hybrid but is potting soil ok for ants because I’m adding in small plants as well such as moss and mini grass patches.

How many workers should I have before I move them in?

Your ants should have access to an outworld and food when the first worker arrives via the drinking straw method. After that it's up to them if they chose to move into the soil.
Proverbs 6:6-8

Go to the ant, you sluggard;
consider its ways and be wise!
It has no commander,
no overseer or ruler,
yet it stores its provisions in summer
and gathers its food at harvest.
