its bacteria, it forms if you or your equipment was not sterilized before hand, make sure you wash your tubes and hands really well when constructing your test tube setup, just make another one and transfer that your colonies

Yes, like ausants did. Tape 2 test tube setups together, shine a light on your original and cover the second. Leave a gap between the two tubes for air. Once the queen has moved, secure he test tube and clean the previous one out.
