Well it’s dependant on the weather not a specific day. But if you found a colony that flew on a certain day one year it’s probably going to fly within a two week period or so as it did the year before. As long as the weather is typical.

Anything can be an outworld. For my test tube outworlds I use small Tupperware containers. You could use an empty butter container for example. You don’t need a big fancy outworld for a test tube. Sounds like she doesn’t like the conditions inside of the nest. Maybe it’s not humid enough. What do you mean the workers are aggressive towards her? They’re trying to drag her back inside the nest? Are you giving them enough food/seeds/sugar water?

Make sure the cable isn’t touching the test tube. And also your ants should have a way out of the test tube/heating area in case it gets too hot.

All test tube set ups will mold eventually. A little bit is fine but a lot can kill the colony. Usually the mold doesn’t seem to cause any problems, but best to move them when it gets bad just in case.
