Hm, are you sure you have attached an image of the setup? You should change the test tubes whenever the cotton gets moldy or if it runs out. The temperature is fine, so you can probably close the light in your closet. The only thing they get out of that is warmth and the feeling of exposure, as ants...

The rubbing alcohol is for the baby powder to stick onto the glass/wall, so it's interesting to see the method prevailing without it. Thanks for the tip.

Either getting acrylic or Ytong doesn't really matter, but Ytong will be easier to hydrate. The size? Carefully dump the jar into the outworld, and cover up the nest space with a cloth to keep it dark. Also hydrate the nest and wait for the workers to discover it!

I'm sure anything like insect-a-slip or any of the barriers mentioned should be alright. If you want to be extra safe, you can just get a lid of any sort (plastic container, etc) and place it over. This will work but it'll most likely kill many ants in the process. Ants might also start to form a bridge with dirt or other pieces of wood that you have provided for them.
