Provide them with water and air, so they do not die. Some queens need to eat while founding a colony, so it may be best to provide them with a drop of honey. If she has dropped her wings off, then she is mated. Otherwise, it is harder to identify, maybe wait and see if she lays eggs.

I have not got any numbers here, but the test tube setup should last a few weeks minimum. It needs to be changed when it runs out of water or when food that you may have put into it rots or causes fungus to grow.
