So I caught I wild spider (where I love there is no way it came to contact with pesticides) froze it and fed it to my camponotus queen and 4 workers in there test tube setup. They have thankfilly accepted it as there forst protien! My question is, how long should I leave it in there? I worry it might cause harmful bacteria to grow

You should not feed them in the test tube, it is very stressful, connect them to a small out-world to forage. I would take it out in a day or two.

Your welcome! A test tube portal would be perfect. :D
