I caught a queen while emptying dirt from plant boxes and i found what I think to be camponotous pencylvannicus or maculatus. before i began digging i saw some workers ontop of the box of what appeared to be the same species. I wasnt able to find any workers or brood though. Will she redo the claustral stage? she is pulling at the cotton a lot.

A claustral queen survives and raises her nanitics by breaking down the flight muscles in her thorax, as well as from proteins from consumption before her nuptial flight. It is unlikely that she will have enough reserves to raise another batch of workers.
You'd be best off releasing her back where you found her.

i found her a worker(from her colony) and gave some food

She still has that one worker with her, she has laid eggs. I am moving her into a new test tube setup, what should i be feeding her?

Feed them broke up insects and sugar water on a little tray so it is easy to clean up

They'll need small, soft-bodied insects and fresh sugar water. Don't move them around unless you absolutely need to (regarding this new test-tube). They will need a foraging area.
