Starting a Myrmica colony

I have recently caught 2 Myrmica Rubra queens and was wondering if this sp. can be raised from a single queen in a test tube or if the queen needs a host colony before she starts laying eggs

Myrmica rubra queens are semi-claustral so you're gonna need to have a small foraging area for them. Also, Myrmica rubra queens are extremely polygynous, so if you're absolutely sure that the queens you have are of the Myrmica rubra species, then you can put those two queens together and they should be fine.
